项目简介和职责:项目组根据中医急性肺损伤理论选取具有清热解毒功效的瑞香狼毒,体内外活性追踪,首次发现瑞香狼毒中全新的黄酮类化合物——狼毒双高色原酮(TT-11)。初步研究发现TT-11在安全无毒的条件下具有显著的抗ALI作用,同时可显著抑制MAPKs活性,减少IL-1β释放。结合文献调研,基于这一新发现,本项目提出“TT-11通过抑制由LPS刺激导致的MAPKs活性,进一步抑制NLPR3的激活,最后使IL-1β的释放减少,发挥抗ALI作用”假说,重点从药效及MAPKs-NLRP3信号轴在ALI的作用入手,从而在细胞和分子水平阐明TT-11抗LPS诱导的ALI的作用及机制。为开发基于中医药理论的抗ALI药物提供理论依据,也为中医药现代化提供新思路和新方法,具有重要的理论意义和应用价值。
